Inhibition of MicroRNA-21 induces apoptosis in dermal fibroblasts of patients with systemic sclerosis

Abstract: Prolonged activation of dermal fibroblasts is the main cause of progressive fibrosis in systemic sclerosis (SSc). It seems that inhibition of apoptosis in SSc fibroblasts deregulates fibrosis. MicroRNA-21 (miR-21) is a pro-fibrotic factor with high expression in lesional areas of SSc skin and fibroblasts. Abstract: Traditional medicines thatstimulateormodulatetheimmunesystemcanbeusedasinnovativeapproachestotreatimmunologicaldiseases.TheherbalmedicineIMODhasbeenshowntostronglymodulateimmuneresponsesinseveralanimalstudiesaswellasinclinicaltrials.However,littleisknownaboutthemechanismsofIMODtomodulateimmunity.HerewehaveinvestigatedwhetherIMODmodulatestheimmunologicalfunctionofhumandendriticcells(DCs).IMODalonedidnotinduceDCmaturationnorproductionofcytokines.Notably,IMODdecreasedtheproductionofpro-inflammatorycytokinesIL-6,IL-12p70,andTNFαbyLPS-activatedDCsatbothmRNAandproteinlevelsinadosedependentmanner.Incontrast,treatmentwithIMODdidnotaffectLPSinduced-productionoftheanti-inflammatorycytokineIL-10.Furthermore,IMODinhibitedTcellactivation/proliferationbyLPS-treatedDCsandskewedT-cellsresponsestowardtheThelpertype2polarization.ThesedatastronglyindicatethatIMODhasapotentimmunomodulatoryabilitythataffectsTLRsignalingandtherebymodulatesDCfunction.InsightintotheimmunomodulatoryeffectofherbalmedicineIMODmayprovideinnovativestrategiestoaffecttheimmunesystemandtohelpcombatvarousdiseases. Delay in myofibroblasts apoptosis results in fibrotic diseases such as systemic sclerosis (SSc). Transforming growth factor-b1 (TGF-b1) is an important cytokine to induce fibroblasts differentiation into myofibroblasts. Abstract: The aim of this study was to assess the role of HLA-B*27 and it’s subtypes in determining severity and clinical manifestations of ankylosing spondylitis (AS). A total of 163 AS patients were assessed for clinical manifestations and severity using structured questionnaires. HLA-B*27 screening and B*27 sub-typing were performed by PCR.
