HerbalmedicineIMODsuppressesLPS-inducedproductionofproinflammatorycytokinesinhumandendriticcells


Abstract: Traditional medicines thatstimulateormodulatetheimmunesystemcanbeusedasinnovativeapproachestotreatimmunologicaldiseases.TheherbalmedicineIMODhasbeenshowntostronglymodulateimmuneresponsesinseveralanimalstudiesaswellasinclinicaltrials.However,littleisknownaboutthemechanismsofIMODtomodulateimmunity.HerewehaveinvestigatedwhetherIMODmodulatestheimmunologicalfunctionofhumandendriticcells(DCs).IMODalonedidnotinduceDCmaturationnorproductionofcytokines.Notably,IMODdecreasedtheproductionofpro-inflammatorycytokinesIL-6,IL-12p70,andTNFαbyLPS-activatedDCsatbothmRNAandproteinlevelsinadosedependentmanner.Incontrast,treatmentwithIMODdidnotaffectLPSinduced-productionoftheanti-inflammatorycytokineIL-10.Furthermore,IMODinhibitedTcellactivation/proliferationbyLPS-treatedDCsandskewedT-cellsresponsestowardtheThelpertype2polarization.ThesedatastronglyindicatethatIMODhasapotentimmunomodulatoryabilitythataffectsTLRsignalingandtherebymodulatesDCfunction.InsightintotheimmunomodulatoryeffectofherbalmedicineIMODmayprovideinnovativestrategiestoaffecttheimmunesystemandtohelpcombatvarousdiseases.
